One egregious omission: Lee Pace as Calpernia Addams in “Soldier’s Girl.” With help of makeup and costuming, he was fairly convincing, and he gave a stellar performance besides that.

John Cameron Mitchell in Hedwig and the Angry Inch. No competition, that movie is pure genius. Plus, the songs are seriously awesome.
What about Tim Curry in Rocky Horror?
Not exactly drag in the sense you mean it here, but how about Billy Crudup in Stage Beauty?
I thought John L. as Chi Chi in To Wong Foo was the 2nd most believable performance, second only to Hillary Swank. He stole the show in that movie. I also love Robin Williams as Mrs. Doubtfire and the mention of John Lithgow and Dustin Hoffman and all three from “Priscilla”. Love that movie!
I’m going to take a guess that none of you have seen Linda Hunt in The Year of Living Dangerously. She plays a small statured man and did win an Academy Award. It wasn’t until the credits that I knew she was a woman in that role.
“I can’t be seen on tv at this present weight!”-Edna Turnblad! John Travolta was amazing in that role, and I actually knew people who thought he really was a woman! lol

Loved Lee Pace in “Soldier’s Girl,” though technically he was playing a MTF transsexual. Such an awesomely powerful performance and movie!!!
I’m so glad Cillian Murphy is ranked most highly, but I’ll second Lee Pace in Soldier’s Girl, as well.
